چکیده
A single 9-amino acid peptide of the defined murine cytotoxic T lymphocyte (CTL) epitope (named peptide-1), which corresponds to the amino acid residues 298-306 (GYISTRVEM) of NS3 of dengue virus serotypes DEN-2 and 4, was examined for induction of specific CTLs. Immunization of BALB/c mice subcutaneously with the peptide-1 emulsified with complete Freund adjuvant (CFA) did not induce specific CTLs. The peptide-2 (GYISTRVEL), in which the residue (M) at 9th position of the peptide-1 was substituted for L, was prepared. The peptide-2 possessed the complete H-2K-binding motif. Intravenous immunization with 5x10 dendritic cells (DCs) pulsed with the peptide-2-induced specific CTLs. Furthermore, subcutaneous immunization with the peptide-2 emulsified with CFA-induced CTLs which lysed peptide-1-pulsed target cells as well as peptide-2-pulsed ones. These results indicate that immunization with dengue virus-derived CTL epitope peptide induces specific CTLs, and that DC can be used as a vehicle for the modified epitope peptide.

Intranasal immunization with cytotoxic T-lymphocyte epitope peptide and mucosal adjuvant cholera toxin: selective augmentation of peptide-presenting dendritic cells in nasal mucosa-associated lymphoid tissue.
We previously reported that cholera toxin (CT) was required as a mucosal adjuvant for the induction of peptide-specific cytotoxic T lymphocytes (CTL) following intranasal immunization with CTL epitope peptides (A. Porgador et al., J. Immunol. 158:834-841, 1997). The present study was performed to identify the site and the antigen-presenting cell (APC) population responsible for the presentation...
متن کاملImmunization with a Lymphocytic Choriomeningitis Virus Peptide Mixed with Heat Shock Protein 70 Results in Protective Antiviral Immunity and Specific Cytotoxic T Lymphocytes
Heat shock proteins (hsp's) isolated from murine cancer cells can elicit protective immunity and specific cytotoxic T lymphocytes (CTLs) by channeling tumor-derived peptides bound to hsp's to the major histocompatibility class I antigen presentation pathway. Here we have investigated if hsp70 can be used in a novel peptide vaccine for the induction of protective antiviral immunity and memory CT...
متن کاملModified vaccinia virus Ankara for delivery of human tyrosinase as melanoma-associated antigen: induction of tyrosinase- and melanoma-specific human leukocyte antigen A*0201-restricted cytotoxic T cells in vitro and in vivo.
Vaccination with tumor-associated antigens is a promising approach for cancer immunotherapy. Because the majority of these antigens are normal self antigens, they may require suitable delivery systems to promote their immunogenicity. A recombinant vector based on the modified vaccinia virus Ankara (MVA) was used for expression of human tyrosinase, a melanoma-specific differentiation antigen, an...
متن کاملA single nine-amino acid peptide induces virus-specific, CD8+ human cytotoxic T lymphocyte clones of heterogeneous serotype specificities
It is generally accepted that virus-specific CD8+ cytotoxic T lymphocytes (CTLs) recognize nine-amino acid peptides in conjunction with HLA class I molecules. We recently reported that dengue virus-specific CD8+ CTLs of two different serotype specificities, which were established by stimulation with dengue virus, recognize a single nine-amino acid peptide of the nonstructural protein NS3 of den...
متن کاملVirosome-mediated delivery of tumor antigen to plasmacytoid dendritic cells.
Cytotoxic T lymphocytes (CTL) are crucial in viral clearance and tumor growth control. Thus the induction of CTL activity is an important aim in vaccine development. We investigate an innovative delivery system for peptide transfer to the MHC class I processing pathway of APC with the aim to trigger CTL in the context of an antitumoral response. The strategy relies on a novel antigen delivery s...
